Abstract

Introduction: Glucagon-like peptide-1 receptor agonists (GLP-1 RAs) are commonly used in patients with type 2 diabetes (T2D) to improve glycemic control and reduce body weight. Tirzepatide is the first dual glucose-dependent insulinotropic polypeptide and GLP-1 RA approved for the management of type 2 diabetes and obesity. In a head-to-head trial, tirzepatide demonstrated significant reductions in mean hemoglobin A1c (HbA1c) and body weight compared to the GLP-1 RA, semaglutide. However, limited data has evaluated the impact of switching to tirzepatide from GLP-1 RAs on glycemic and weight parameters.

Research Question or Hypothesis: Does switching to tirzepatide from GLP-1 RAs improve glycemic and weight parameters?

Study Design: Single center, retrospective pre-post cohort study

Methods: Adults with T2D initiated on tirzepatide between June 2022 to May 2024 and receiving a GLP-1 RA at the time on initiation will be included. Patients must also have at least one follow-up visit within 6 months of tirzepatide initiation. Patients will be excluded if they are pregnant or not receiving GLP-1 RA within 2 weeks of initiation. An initial report has been generated through the clinic’s electronic health record based on the inclusion criteria to capture eligible patients. Two researchers will complete chart reviews to identify eligible patients and collect data. The primary endpoint will be changes in HbA1c from baseline to 3 and 6 months. Secondary endpoints include change from baseline body weight to follow-up and occurrence of adverse effects. Paired t-tests will be used to assess average changes from baseline to follow-up.

Results: A total of 279 patients will be screened. Results are pending.

Conclusion: Results are pending.
